Novel synergism of two antifungal agents, copiamycin and imidazole.

Copiamycin, a macrocyclic lactone antifungal antibiotic, was found to potentiate the antifungal effect of imidazole compounds, ketoconazole in particular. The potentiation of two nominally fungistatic agents in vitro was substantiated by a marked reduction of the minimum inhibitory and minimum fungicidal concentrations when the drugs were used in combination. The effectiveness of this synergist...

Potentiation of rifampicin, rifampicin analogs, and tetracycline against animal cells by amphotericin B and polymyxin B.

A number of antibiotics alter the permeability of the membranes of eukaryotic cells (1). Previously, we have exploited this property of amphotericin B to increase the uptake of 2nd drugs into fungi and animal cells, thereby enhancing the effects of the 2nd agents (5—7). Recently our studies have been confirmed by Kuwano et al. (4). This report is an extension of our previous work and represen...

In vitro susceptibilities of sucrose-negative Candida tropicalis, Candida lusitaniae, and Candida norvegensis to amphotericin B, 5-fluorocytosine, miconazole, and ketoconazole.

The MICs and minimal lethal concentrations of four antimycotics, amphotericin B, 5-fluorocytosine, miconazole nitrate, and ketoconazole, were determined for 25 yeast isolates representing species uncommonly implicated in candidiasis. A microdilution procedure was employed with complex and synthetic media. The isolates, in general, were susceptible to the same antimicrobial agents shown to be ef...

Rifampin: inhibition of ribonucleic acid synthesis after potentiation by amphotericin B in Saccharomyces cerevisiae.

The inhibition of Saccharomyces cerevisiae growth by amphotericin B and rifampin was studied. Rifampin alone had no effect on growth or macromolecular syntheses. Lethal amounts of amphotericin B produced a late inhibition of ribonucleic acid (RNA) synthesis simultaneous with the arrest of growth and protein synthesis. In contrast, low doses of amphotericin B along with rifampin caused an early ...

Activity of amphotericin B, 5-fluorocytosine, and rifampin against six clinical isolates of Aspergillus.

Amphotericin B in combination with 5-fluorocytosine was synergistic against three clinical isolates of Aspergillus fumigatus and one of three clinical isolates of A. flavus. Amphotericin B in combination with rifampin was synergistic against all six clinical isolates of Aspergillus tested. The levels of 5-fluorocytosine and rifampin required for synergism were higher than clinically achievable ...
